摘 要:目的探讨大蒜素对K562细胞株及裸鼠移植瘤的作用。方法利用MTT法检测不同浓度大蒜素对K562细胞的增殖抑制影响;Annexin V-FITC标记法检测K562细胞凋亡;建立K562细胞Balb/c裸鼠皮下移植瘤模型,荷瘤小鼠随机分成4组,分别用1.5、3、6mg/ml大蒜素和生理盐水对瘤体进行局部注射,每次0.1ml,隔日1次,连续7次,观察瘤体变化。结果不同浓度大蒜素对K562细胞均有抑制作用,呈明显的剂量依赖性,抑制率分别为21.5%、55.1%、81.6%(P<0.05);0.005mg/ml大蒜素诱导K562细胞凋亡作用最显著,0.025mg/ml大蒜素使K562细胞呈中毒反应,凋亡作用不明显;大蒜素组瘤体生长较对照组明显受到抑制,抑瘤率分别为18.5%、79.2%、91.6%。结论大蒜素具有显著抑制K562细胞增殖及促凋亡的作用,能有效抑制K562细胞Balb/c裸鼠移植瘤的生长。Objective To study the anti-tumor effect of allicin on human leukaemia cancer cell line K562 and Balb/c nude mice tumor xenograft. Methods Cltural cell lines of human leukaemia cancer K562 were used as the target cells to observe the effect of allicin of different concertritions on these cell lines by MTT assay. Detecting the quantitation of cells apoptosis by Annexin V-FITC marking method. The transplanted tumor model was established by injecting human leukaemia cancer cell line K562 into subcutaneous tissue of nude mice. Tumor-bearing mice were divided into 4 grougs at random:0.1 ml of normal saline solution, 1.5 mg/ml allicin,3 mg/ml allicin,6 mg/ml allicin directly injected into tumor body respectively, for seven times at an interval of two days to observe the changes of tumor volume, the tumor inhibiting rate. Results Allicin showed significant inhibiting effect on the growth K562 cells,exhibiting a dose-dependent relationship,the inhibiting rates were 21.5% ,55.1% and 81.6% respectively ( P 〈 0.05 ). The apoptotic rate of cells was the best significantly when the concentration of allicin was 0.005 mg/ml in 24 hours. In the groups of allicin intervention, it showed a marked delay in the appearance and growth of tumors after injection compared with control group,the inhibiting rates were 18.5% ,79.2% and 91.6% respectively. Conclusion Allicin can inhibit the growth of K562 cells and induce the apoptosis. Allicin can also inhibit the growth of transplanted K562 cells effectively in vivo by interventional method.
