摘　　要：目的:探讨Neuropilin-1(Nrp-1,神经菌毛素)在小鼠黑色素瘤细胞体外增殖中的作用。方法:用活细胞荧光染料羧基荧光素乙酰乙酸琥珀酰亚胺酯(carboxyfluorescein diacetate succinimidyl ester,CFDA-SE)标记小鼠B16-F10黑色素瘤细胞后,将肿瘤细胞分别用Nrp-1阻断抗体及Nrp-1配体血管内皮生长因子165(vascular endothelial growth factor165,VEGF165)阻断抗体预孵育,体外培养72小时后流式细胞术结合Modfit软件分析肿瘤细胞增殖情况。结果:CFDA-SE标记黑色素瘤细胞生长良好,非抗体阻断组的肿瘤细胞增殖指数显著高于Nrp-1抗体阻断组[(20.67±1.16)%vs(12.67±0.28)%,P<0.01]及VEGF-165抗体阻断组[(20.67±1.16)%vs(15.15±0.83)%,P<0.01],两个抗体阻断组的增殖指数差异也非常显著(P<0.01)。结论:Nrp-1及其配体VDGF-165对小鼠黑色素瘤细胞的增殖有显著地促进作用,阻断Nrp-1产生的抑制肿瘤细胞增殖作用比阻断VEGF165更强;VEGF165/Nrp1直接作用及Nrp-1/Nrp-1直接作用也可能是肿瘤细胞增殖的重要机制。Objective： To verificate the effect of Neuropilin - 1 （ Nrp - 1 ） in proliferation of mice melanoma cells. Methods：After labelled by carboxyfluorescein diacetate succinimidyl ester（ CFDA -SE）, melanoma cells were preincubated with Nrp - 1 blocking antibody and vascular endothelial growth factor165 （ VEGF165 ） blocking antibody and were cultured in vitro for 72h. Flow cytometry and Modifit soft were used to analyse the proliferation of melanoma cells. Results： Effect of CFDA -SE labelling was excellent. Proliferation index（PI） of the group without blocking antibody was significantly higher than that of the group withNrp - 1 and VEGF165 blocking antibody [ （20.67 ± 1.16）% vs （12.67±0.28）% ,P〈0. 01 and [ （20.67 ±1.16）% vs （15.15±0. 83）% ,P〈0.01]. There was also significant statistical difference between the two groups of blocking antibody [ （ 12.67 ±0.28 ） % vs （ 15.15±0.83）%,P 〈 0. 01 ]. Conclusion： Nrp- 1 and its ligand VEGF165 have important promotion effect on prolifera- tion of mice melanoma cells. Blocking Nrp - 1 is more effective than blocking VEGFI65 in inhibitting the proliferation. Derect mutual effect of melanoma cells through Nrp - 1/VEGF165 or Nrp - 1/Nrp - 1 pathway may also be important mechanisms in tumor cells proliferation.
