摘 要:[目的]研究红曲霉转化中药葛根固体发酵的条件,以期提高其转化率。[方法]对菌株YM3207转化葛根干粉的固体发酵培养条件和培养基进行了优化试验,通过菌种活化、种子培养、固体发酵培养方法以及正交试验考察了种子培养基、底物投入量、转化温度、转化时间因素的影响与红曲霉转化条件。[结果]红曲霉菌YM3207菌株在YPD液体培养基中28℃、200r/min下培养3d作为种子培养条件最为合适;采用固体发酵方式,以中药葛根为底物,10%接种量,28℃培养10d为红曲霉菌最佳转化条件;培养基组成为葛根粉:鲜豆渣:米粉为1:1:1,含水量为50%时,能够获得较高葛根素含量的转化产物。[结论]采用该条件发酵转化葛根能够提高葛根主要有效成分含量,利用微生物转化提高中药药效是可行的。[Objective]The aim was to research the condition on solid fermentation conditions of Monascus purpureus for transforming Chinese medicine Puerariae radix in order to increase the conversion rate.[Method]The solid fermentation culture conditions and the medium of strain YM3207 for transforming the dry powder of Puerariae radix were made for the optimization test.Through the strain activation,seed culture,solid fermentation culture method and orthogonal test,the effects of some factors such as the seed medium,substrate input amount,transition temperature,transition time factors and the transition condition of Monascus purpureus were investigated.[Result]The seed condition when the strain of Monascus purpureus YM3207 was cultured in the YPD liquid medium under 28 ℃ and 200 r/min for 3 d was optimum.The optimum transition condition of Monascus purpureus strain was as follows:taking Chinese medicine Puerariae radix as the substrate with inoculation amount of 10% and culturing for 10 d at 28 ℃ by using the solid fermentation method.When the medium compositions was Puerariae radix powder,fresh bean cake and rice flour with ratio of 1:1:1 and the water content as 50%,the transition product with higher Puerariae radix content was obtained.[Conclusion]Using this fermentation condition for transforming Puerariae radix could increase the mainly effective composition content of Puerariae radix and it was feasible to transform and increase Chinese medicine by using microorganism.
