摘 要:应用Plackett-Burman设计法对影响乳酸乳球菌发酵的培养基主要组分进行筛选,确定了影响乳链菌肽效价的关键因素为酵母浸粉、葡萄糖和K2HPO4.在此基础上,采用响应面法(RSM)优化乳链菌肽发酵培养基.结果表明,当酵母浸粉质量浓度为22.35g/L,葡萄糖质量浓度为24.80g/L,K2HPO4质量浓度为13.65g/L时,乳链菌肽产量最大,此时,乳链菌肽产率为2157IU/mL.实验验证,最佳条件下乳链菌肽产率为2115IU/mL,预测值与验证值吻合得较好.The Plackett-Burman design was used to evaluate the effects of eight variables which could affect the nisin production. On the basis of analyzing the statistical regression, yeast extract, glucose and K2HPO4 were found to be the most important factors. Subsequently, response surface methodology was employed to determine the maximum nisin production at the optimum concentration of key factors. The nisin production was found to correlate to the three parameters. The optimal values of the three parameters were determined as 22.35 g/L yeast extract, 24.80 g/L glucose and 13.65 g/L K2HPO4. The nisin production was 2 157 IU/mL, and the actual experimental result was 2 115 IU/mL, which shows the results are reasonable.
