摘 要:以香椿叶多糖提取率(Y1)和香椿叶多糖含量(Y2)为响应值,利用响应面法对香椿叶多糖提取工艺进行多目标同步优化。根据统计模型发现液料比、提取时间、提取温度3个因素均对香椿叶多糖提取率和多糖含量有显著影响。由响应面三维图及等高线叠加图推测得到香椿叶多糖提取率与含量均较高的最佳提取工艺参数为液料比40:1(mL/g)、提取时间3.1h、提取温度75℃。验证优化工艺参数得到多糖提取率为4.75%、多糖含量为58.5%,与模型预测值非常接近,采用响应面法对香椿叶多糖提取条件进行优化合理可行。Toona sinensis (A.Juss.) Roem leaves were used as the raw material to extract polysaccharides by hot water extraction.Response surface methodology (RSM) was employed to maximize polysaccharide yield (Y1) and purity (Y2) based on a central composite design involving 3 factors (liquid/solid ratio,extraction time and temperature) at 3 levels.Results showed that all 3 factors had significant effects on Y1 and Y2,and their optimal values were determined with three-dimensional response surface diagrams and overlaying contour plots as follows:liquid/solid ratio 40:1,extraction time 3.1 h,and temperature 75 ℃.Under such conditions,the observed values of Y1 and Y2 were 4.75% and 58.5%,respectively and were in close agreement with the predicted values.This reveals the applicability of RSM to the optimization of extraction of polysaccharides from Toona sinensis (A.Juss.) Roem leaves.
