摘 要:目的:研究肺癌MDM2蛋白表达与p53突变之间的关系。方法:用免疫组化检测125例肺癌p53和MDM2蛋白表达,以聚合酶链反应单链构象多态性(PCRSSCP)方法检测p53基因第5~8外显子的突变。结果:PCRSSCP检测阳性的52例肺癌MDM2阳性率为19.2%;而阴性的73例肺癌MDM2蛋白阳性率为35.6%。统计学检验显示肺癌MDM2蛋白表达与p53突变之间呈负相关(χ2=3.98,P<0.05);MDM2蛋白阳性与肺癌患者的年龄、性别、肿瘤大小、组织学类型、分化程度、临床分期和p53蛋白表达无显著相关性。Objective: To study the correlation between MDM2 overexpression and p53 mutation in lung cancer. Methods: Immunohistochemistry and nonisotopic PCR SSCP were used. Results: The positive rate of MDM2 protein in 52 cases with SSCP positive was 19.2%, and in 73 cases with SSCP negative was 35.6%. In statistics, there was a negative correlation between MDM2 expression and p53 mutation (χ 2=3.98, P <0.05). The MDM2 expression status in human lung cancer was not associated with age, sex, clinical stage, histological subtypes, tumor sizes, lymphatic metastasis, and prognosis. Conclusion: MDM2 might play an important role in the development of the lung cancer without p53 mutations.
