摘  要:在人体生理酸度(pH7.4)条件下,运用荧光和紫外光谱法研究了红景天苷与人血清白蛋白(HSA)结合作用的光谱特性。结果显示,随着红景天苷浓度的增大,HSA的荧光强度明显增强并且发生蓝移;HSA-红景天苷复合物与红景天苷的紫外差谱及逐渐增大的荧光各向异性,进一步说明红景天苷能够与HSA发生结合作用。由相关荧光数据求得不同温度下HSA-红景天苷复合物的结合常数及结合反应的热力学参数,推断出二者主要是靠氢键和范德华力结合。同步荧光和三维荧光光谱表明,红景天苷与HSA结合后对HSA构象产生了影响。The spectral characters of the interaction between salidroside and human serum albumin (HSA) were investigated by fluorescence and UV-VlS absorption spectroscopy in physiological buffer solution (pH7.4).The experimental results showed that the fluorescence intensity of HSA could be highly enhanced accompanied with hypsochromic shift with the addition of salidroside.The difference absorption spectra between HSA-salidroside and salidroside at the same concentration and gradually increase in fluorescence anisotropy (r) of salidroside further indicated that salidroside could interact with HSA in solution.The binding constants were obtained at three different temperatures.The calculated thermodynamic parameters from the relevant fluorescence data suggested that the interaction between salidroside and HSA was driven mainly by hydrogen bonds and van der Waals force. The results of synchronous fluorescence spectra and three-dimensional fluorescence spectra demonstrated that salidroside induced a conformational change of HSA.
