摘 要:采用温差并结合超声波辅助的复合花粉破壁法对花粉进行破壁,先对温差法进行正交试验L9(34),得到的最佳工艺条件为温差100℃、水浴时间7h、料液比1:10、冷冻时间24h,在此条件下,花粉的破壁率为61.61%。接着对温差处理过的样品继续进行超声辅助破壁工艺研究,采用正交试验L9(33),在功率为200W的情况下,得到最佳工艺为超声温度60℃、时间30min、浓度5%,此时花粉的破壁率达到88.53%。The method of wall-breaking by temperature difference and ultrasonic were used in pollen.The wall-breaking method by temperature difference was optimized with L9(3^4) orthogonal test,optimum wallbreaking conditions were determined as follows:temperature difference 100 ℃,time of water-bath 7 h,ratio of material to liquid 1:10 and refrigerant time 24 h,under these conditions,the wall-breaking rate is 61.61%.And then carrying out wall-breaking by ultrasonic,experiments were made by using L9(3^3) orthogonal test,when ultrasonic power is 200 W,optimum conditions were:ultrasonic temperature 60 ℃,ultrasonic time 30 min and liquid concentration 5%,the wall-breaking rate reached to 88.53% under these conditions.
