表达α-2a干扰素工程菌的大型罐级联控制溶氧发酵研究

摘　　要：用500L大型发酵罐级联控制溶氧培养表达α－2a干扰素的大肠杆菌，获得了菌体生长与培养时间呈直线关系的结果（b＝6．668，r＝0．9963，P＜0．01）。多批实验结果证明，菌体单产（g／L）为36.148±2.13，菌体罐产（g／罐）为11613±210.06，IFN活性为5.086±2.93（×108Iu／L）。The recombinant E. colt expressing IFN-α2a was cultured in 500L fermenterunder the condition of cascade controlling dissolved oxygen content. The result showed alinear relation between cell density and culture time (b= 6. 6668, r= 0. 9963, P<0.01). The celloutput per liter and per fermenter were 36.148±2. 13g and 11613±21. 06g respectively, andthe activity of IFN was 5. 086 × 108±2. 93 ± 108IU/L.
