摘  要:目的通过检测LRIG1在胶质瘤细胞中的亚细胞定位及LRIG1对EGFR的作用,探讨LRIG1对EGFR信号通路的抑制效应。方法用Confocal法及Westernblot法检测LRIG1质粒转染前后H4细胞中EGFR和LRIG1表达,MTT法检测LRIG1对细胞增殖的影响。结果在H4细胞中LRIG1低表达,EGFR高表达,均主要为膜表达;转染LRIG1质粒后,LRIG1表达增强,EGFR表达下降;细胞的生长受抑制。结论LRIG1主要位于胶质瘤细胞膜上,通过促进EGF受体的降解抑制其信号传导。Objective To explore the LRIG1 inhibition effect on EGFR signaling by deteimining the subcellular localization of LRIG1 and its effect on EGFR in the glioma cells. Method The expression of EGFR and LRIG1 in H4 cells before and after LRIG1 plasmid transfeetion were determined by Confocal and Western blot assays. The impact of LRIG1 on cell growth was analyzed by MTT. Results The expression level of LRIG1 was low and EGFR was high in H4 cells. They were mainly expressed on the plasma membranes. After transfection, LRIG1 was upregulated, while EGFR was downregulated. The growth of cells was inhibited. Conclusions LRIG1 is mainly located on the cell membrane, which impairs EGFR downstream signaling by enhancing degradation of the receptors.
