摘 要:为比较鸡Ⅹ期胚盘细胞分散培养与整胚培养效果,对鸡Ⅹ期胚盘细胞分别进行分散培养和整胚培养并进行传代,观察细胞生长情况,通过碱性磷酸酶(AKP)染色鉴定计算阳性克隆率;结果,分散培养的细胞生长状态良好,整胚培养的细胞贴壁能力较差,培养过程中细胞易分化;传代后两种方法培养的细胞AKP阳性克隆率差异显著(P<0.01)。表明鸡Ⅹ期胚盘细胞分散培养效果较为理想。To contrast the effect of chicken blastodermal cells on X period respectively by scattered culture and the whole embryo culture,the chicken blastodermal cells on X period were separated cultured with two methods. The growth of cells was observed carefully. The rate of positive clones was calculated by ceils subcuhured and identified with AKP staining. The growth of separated cultured cells were in good condition and the whole embryo cultured cells showed weaker adherent ability and were easier to be differentiated. There was significant difference between the two cultural methods. The result of chicken blastodermal ceils on X period cultured by scattered culture was better.
