摘 要:目的研究还原叶酸载体基因(Reduced folate carrier,RFC1)A80G多态性与儿童急性淋巴细胞白血病(Acute lymphoblastic leukemia,ALL)易感性的关系。方法本研究为病例对照研究,采用DNA直接测序法对30例ALL儿童(ALL组)及70名健康儿童(对照组)进行RFCl80A→G基因型的测定,分析该基因型在ALL组与对照组之间的分布差异。结果与携带80GG基因型者比较,携带80AA基因型者ALL风险增加4.5倍(OR=4.50,95%CI:1.17-17.30);以携带80GG和GA基因型者为参照,80AA变异基因型者ALL风险增加3.22倍(OR=3.22,95%CI:1.24-8.41)。结论RFC1A80G多态可能是儿童急性白血病发病的易感因素之一。Objective To study the relationship between polymorphisrn of reduced folate carrier gene RFC1-A80G and the susceptibility of childhood acute lymphoblastic leukemia (ALL). Methods A case-control study was performed. DNA sequencing was applied to detect the genotype polymorphism of RFC1-A80G in 30 children with ALL and 70 healthy children as the control group. The distribution difference of the genotype between them was analyzed. Results Logistic regression analysis revealed that individuals with 80AA wild genotype had a significantly increased risk (4.50 fold) of ALL (OR =4.50; 95% CI: 1.17-17.30), compared with 80GG variant genotype. Furthermore, individuals with 80AA genotype had a 3.22-fold risk of ALL ( OR = 3.22; 95% CI: 1.24-8.41 ) compared with 80GG/GA genotypes. Conclusion The polymorphism of RFC1-A80G gene may be one of the potential susceptible factors in leukemogenesis for Chinese children.
