摘  要:本文以0.25mol/L的NaOH-乙醇溶液作提取剂,从核桃仁种皮中提取多糖,提取液经去脂、去蛋白、醇析后得粗多糖WKPP-70。利用DEAE-Sephadex A-25凝胶柱和Sephadex G-75凝胶柱对粗多糖进行分离纯化,得纯多糖WKPP-70-1-1。通过紫外光谱分析证明其纯度,红外光谱分析显示其具有多糖类的特征吸收峰。抑菌试验表明,WKPP-70-1-1对大肠杆菌、沙门氏菌、枯草芽孢杆菌、金黄葡萄球菌均有一定的抑制作用。Crude polysaccharide, WKPP-70, was extracted from walnut kernel pellicle, using 0.25 mol/L NaOH-ethanol. After removal of lipids and proteins, WKPP-70 was further purified by chromatography on DEAE-Sephadex A-25 and Sephadex G-75 columns. The purity of the obtained polysaccharide, named as WKPP-70-1-1 was determined using UV spectrophotometric method and its structure was analyzed by infrared spectrum (IR). The antimicrobial activities of WKPP-70-1-1 against E.scherichia coli, Bacillussubtilis, Staphy lococcus aureus and SalmonellaTyphimurium were also evaluated. The results showed that WKPP-70-1-1 had antimicrobial activities against these examined microorganisms.
