10-HDA对原代培养大鼠海马神经元增殖的研究

摘　　要：研究10-HDA对原代培养新生大鼠海马神经元的增殖效果.原代培养24 h的新生大鼠海马神经元,通过添加不同浓度的10-HDA(0.1μmol/L、1.0μmol/L、10μmol/L)作用后,利用BrdU免疫荧光染色检测神经细胞增殖,尼氏染色观察神经元数量的变化.结果显示中剂量浓度(1.0μmol/L)的10-HDA作用海马神经元24h后,细胞数量明显增多;而高浓度(10μmol/L)10-HDA作用海马神经元24 h后,细胞数量明显低于正常对照组和低剂量组(0.1μmol/L).BrdU免疫荧光染色表明,中剂量浓度的10-HDA可使细胞增殖,增殖率达到(19.37±2.192 1)%.尼氏染色结果表明,10-HDA使海马神经元数量增加了(20.31±4.086 5)%.研究结果表明,10-HDA对原代培养的海马神经元有增殖作用.To observe the multiplication effect of primary cultured hippocampal neurons of new born rats, the primary cultured postnatal rat hippocampal neurons was added different concentration of 10-HDA（0.1, 1.0,10 μmol/L）, then neural cell proliferation was checked by Brdu immunofluorescence staining. Nissl staining was used to observe the quantitative change of neurons. T, he results shown that the quantity of hippocampal neurons added medium dose 10-HDA （1.0μmol/L） increase obviously after 24 hours, but the quantity of hippocampal neurons added 10/μmol/L 10-HDA was lower than the control group and the lowdose group（0. 1 μmol/L）. Brdu immunofluorescence staining shows that medium dose 10-HDA make cells proliferation,growth rate is（19.37±2. 192 1）% ,Nissl staining shows 10-HDA can increase the amount of hippocampal neurons, rowth rate is （20.31 ± 4. 086 5）%. 10-HDA can promote the proliferation of primary cultured hippocampal neurons.
