摘　　要：目的研究Aurora-A高表达对人食管癌细胞的影响,以了解其在人食管癌侵袭转移中的作用。方法采用细胞转染的方法,利用Aurora-A基础水平表达较低的食管癌细胞系构建Aurora-A高表达细胞株,比较其与转染空载的细胞,分析二者在表型差别。结果Aurora-A高表达细胞表现出明显的增殖优势,在体外细胞移动实验中表现出穿膜能力大大增强,致瘤作用明显增强;提示Aurora-A高表达在人食管癌侵袭转移中有重要的意义。Objective Observing the effect of Aurora-A on the esophageal cancer cell lines in order to analyze its role in malignancy development of human esophageal cancer.Methods We developed the Aurora-A overexpressing cell lines by transfecting this gene into the esophageal cancer cells that have low basal level of Aurora-A and then investigated the phenotypic alterations of these cell lines.Results With increased expression of the Aurora-A protein,the cells exhibit faster proliferation and stronger motility ability.Conclusion Aurora-A may contribute to invasive growth and metastasis of human esophagus cancer by promoting cell proliferating and motility.
