摘 要:探讨粪鬼伞多糖提取过程中的脱蛋白方法与工艺。以蛋白质去除率和多糖损失率为考察指标,比较Sevag法、酶法、三氯乙酸法、酶-Sevag法脱蛋白效果。结果表明,酶-Sevag法脱蛋白效果较好且多糖得率较高,脱蛋白的最佳工艺条件为:酶用量2%,温度40℃,脱蛋白时间2h,pH值5.0;Sevag试剂浓度20%,脱蛋白次数3次,在此条件下,蛋白质去除率和多糖损失率分别为70.95%和6.58%。Investigated the method of deproteinization in Coprinus sterqulinus Fr. polysaccharide extraction. The effect of deproteinization with different methods(such as Sevag, enzyme, TCA and enzyme-Sevag method)were evaluated in terms of the ratio of protein removal and polysaccharide loss. The result showed that the enzyme-Sevag method could be the best, with less protein remain and polysaccharide loss. The optimum technological conditions of deproteinization were: enzyme usage 2%, temperature at 40 ℃, pH value 5.0, deproteinization time 2 h, Sevag concentration 20%, deproteinization times 3 times. The ratio of deproteinization and polysaccharide loss were 70.95% and 6.58%, respectively. Conclusion:Enzyme-Sevag method could be used for Coprinus sterqulinus Fr. polysaccharide deproteinization.
