摘  要:家蚕茧壳经过高温高压水脱胶处理后,被覆在丝纤维外面的绝大部分丝胶层物质(色素、蜡质和碳水化合物等)都溶解在水中。利用乙醇溶液反复沉淀蛋白的方法,分离和纯化了5种不同蚕茧品种的丝胶蛋白(SDS-PAGE分析表明这些蛋白分子量都分布在10~100 kDa之间),对已纯化丝胶样品的体外生物活性如抗氧化、抑制酪氨酸酶活性和抑制α-葡萄糖糖苷酶活性进行分析。结果表明,分离纯化丝胶蛋白均具有一定的清除自由基、抑制黑色素生成和抑制α-葡萄糖苷酶活性的作用,但生物活性不存在明显的品种间差异。可广泛用于高级化妆品,也可作为抗氧化保健食品和糖尿病降血糖的辅助药物或保健品。When silk fiber derived from Bombyx mori cocoon were subjected to degumming treatments in water under high-temperature and high-pressure, most of the material coating on silk fibers (including pigments, waxes, and carbohydrates) were dissolved in water. Using the method of ethanol precipitated protein repeatedly, five kinds of purified silk sericin samples were obtained(SDS-PAGE analysis showed that the molecular weights of these proteins were distributed between 10-100 kDa). The bioactivities of antioxidation, anti-tyrosinase and anti-α-glucosidase of those purified seriein samples were determined in vitro. The results showed that these samples had activities of scavenging free radicals, inhibiting the production of melanin and anti-α-glucosidase activity. However, there was no significant difference among these species. Because of the anti-oxidant activity, whitening effect and anti-α-glucosidase activity, they can be not only used in cosmetics, but also applied to secondary drugs which suppress diabetes postprandial blood glucose increase and health foods.
