摘  要:目的:探讨核转录因子kB(NF-kB)在牙龈成纤维细胞(GF)中活性的变化及其在牙周病发生中的意义,探讨NF-kB在牙周病调控中的作用及途径。方法:采用免疫细胞化学染色法,观察NF-kB在GF中活性的变化。结果:以10μg/mL的LPS作为有效刺激浓度刺激GF,6h后NF-kB核转位达峰值;以10~100μg/mL浓度的LPS作用GF时,NF-KB激活程度不同,表明10~100μg/mL浓度的LPS是NF-kB激活的有效浓度。结论:健康牙龈组织GF细胞中NF-KB处于非激活状态,LPS可诱导人GF细胞中NF-kB激活发生核转位,LPS通过调控NF-KB的激活调节炎症因子对牙周组织的破坏。Objective:To investigate the regulatory mechanism of nuclear transcription factor-kB(NF-kB) in periodontal disease by observing the lipopolysaccharide to NF-kB activity in human gingival fibrocytes(GF) and its significant to periodontal diseases.Method:The changes of NF-kB activity in human GF were observed with immunocytochemical technique.Result:After being stimulated by effective concentration of LPS with 10μg/ml,nucleus translocation of partial NFkB occurred at 3 h.The peak was at 6h.Concentration of 10 μg/mL~100 μg/mL was probably effective concentration of LPS to stimulate the cells and presented the different activity.Conclusion:Nucleus translocation of NF-kB could occur in cultured human GF induced by LPS,the NF-kB pathway could make it possible for LPS to induce GF to synthesize and secrete the pathogenesis factors.
