摘  要:目的 探讨中国湖南汉族人群FGA-128C/G基因多态性与脑梗死的关系及其对血浆纤维蛋白原(fibrinogen,Fg)的影响.方法 应用聚合酶链反应-限制性酶切片段长度多态性方法对中国湖南汉族194例脑梗死患者和114名与之年龄、性别等相匹配的健康体检者进行FGA-128C/G基因多态分析,并经基因测序证实.结果 在中国湖南汉族人群中,FGA-128C/G存在CC、CG基因型,未发现GG基因型;两种基因型和等位基因频率在脑梗死组和对照组之间比较差异无统计学意义(P〉0.05);Fg水平在两种基因型之间比较差异亦无统计学意义(P〉0.05);通过对血浆Fg影响因素的多元回归分析发现,Fg水平与FGA-128C/G基因型无关(P〉0.05),而随年龄增加而升高,男性高于女性(回归系数分别为0.079,-0.534).结论 中国湖南汉族人群存在FGA-128C/G基因多态性;FGA-128C/G基因多态性与脑梗死患者血浆Fg水平增高无显著相关性,可能不是湖南汉族人群脑梗死的遗传易感基因;年龄和性别是湖南汉族人血浆Fg水平的主要影响因素.Objective To investigate the relationship of FGA gene 128C/G polymorphism and cerebral infarction (CD and evaluate the effect of FGA- 128C/G polymorphism on plasma fibrinogen in Hunan Hans. Methods FGA -128C/G polymorphism was detected by (polymerase chian reaction-restrction fragment length polymorphism, PCR-RFLP) and DNA sequencing in 194 CI patients and 114 healthy controls. Results There were CG and CC genotypes in the FGA- 128C/G locus. No GG genotype was observed in Hunan Hans. There was no significant difference in genotype and allele frequencies between the controls and CI group (P〉0. 05), and statistically significant difference was not found in fibrinogen (Fg) level between the CG and CC genotypes (P〉0. 05). After analyzing blood plasma Fg using the influencing factor multiple regression analysis, it was shown that the Fg level had no relationship with the FGA- 128C/ G genotype, but it increased with age. And the Fg level in males was higher than that in females. Conclusion There was FGA gene 128C/G polymorphism in the Hunan Han population. There was no association of this polymorphism with the increased Fg level of CI patient in the population. FGA-128C/G might not be the predisposing gene of CI in Hunan Han population. The age and sex were the major factors affecting the plasma Fg level in this population.
