摘 要:目的:探究天然植物成分(柠檬提取物)对变形链球菌葡糖基转移酶和细胞外多糖合成的影响。方法:采用二倍稀释法,用含2%蔗糖的胰蛋白胨大豆肉汤(TSB)将柠檬提取物稀释为从1.250μl/ml到0.039μl/ml共计6个浓度的溶液,以TSB液体培养基作为阴性对照组。加入变形链球菌菌液,采用Neson-Somogyi法测定培养6、18、24、48h培养物中的还原糖含量,计算出酶活性;同时采用蒽酮法测定培养6、18、24、48h培养物中的水不溶性多糖和水溶性多糖的含量。结果:随着柠檬提取物溶液浓度的升高,葡糖基转移酶活性和细胞外多糖含量逐渐降低,在各个时间点,从0.078μl/ml组到1.250μl/ml组,各实验组总体均数间均具有显著性差异(P<0.01),各实验组与对照组酶活性、水不溶性多糖含量和水溶性多糖含量都有显著性差异(P<0.01);葡糖基转移酶活性和水不溶性多糖含量之间呈正相关关系(r=0.825~0.963,P<0.01),葡糖基转移酶活性和水溶性多糖含量之间亦呈正相关关系(r=0.815~0.950,P<0.01),且葡糖基转移酶活性与水不溶性多糖含量、水溶性多糖含量的相关性具有显著性差异(t=4.37,P<0.01)。结论:柠檬提取物对变形链球菌葡糖基转移酶和细胞外多糖的合成具有显著抑制作用。Objective: To study the effect of lemon peel extracts on glucosyhransferase and extracellular synthesis of gluean of Streptococcus mutans. Methods: The lemon peel extracts was used as the experimental group with concentrations ranging from 1.250 p^l/ml to 0.039 pJ/ml prepared with Trypticase Soy Broth (TSB) medium with contained 2% glucose, and TSB culture medium was used as the control group. Streptococcus mutans was added to each group ,and cultured for 6, 18, 24, 48 hours by Neson- Somogyi method to measure the content of reducing sugar,and enzyme activity was alculated. The content of water- insoluble glucan and water- soluble glucan were measured by anthrone method. The data was statistically analyzed by one-way ANOVA using SAS9.0 software package. Results: The glucosyltransferase activity and the concentration of extra-cellular glucan decreased gradually with the increase of concentration of lemon peel extracts,and there were highly significant differences (P〈0.01) in the groups of 0.078ul/ml with 1.250 ul/ml lemon peel extracts. There were highly significant differences of extra-cellular gluean between each experimental group and control group (P〈0.01), and between glucosyhransferase activity or specific activity of each experimental group and control group as well (P〈0.01).There was positive correlation between glucosyhransferase activity and water- insoluble glucan content (r=0.825 -0.963 P〈0.01 ),and between glueosyltransferase activity and water- soluble gluean content as well(r=0.815-0.950, P〈0.01), and there were highly significant differences of two correlations (t=4.37, P〈0.01 ). Conclusion: Lemon peel extracts could inhibit the glucosyhransferase activity, extra- cellular synthesis of water- insoluble and water- soluble glucan of Streptococcus mutans.
