摘 要:目的探讨苯巴比妥对癫大鼠海马形态和Bcl-2、Bax蛋白表达的影响。方法取4周龄SD大鼠84只,随机分为4组:9 g.L-1盐水组(A组,18只)、苯巴比妥组(B组,18只)、致后9 g.L-1盐水组(C组,24只)和致后苯巴比妥组(D组,24只)。C组、D组大鼠连续5 d腹腔注射戊四氮(PTZ)60 mg.kg-1.d-1,制造癫模型。第6天起,A组、C组大鼠腹腔注射9 g.L-1盐水2 mL,B组、D组大鼠腹腔注射苯巴比妥50 mg.kg-1.d-1。于喂药9 d、15 d、21 d,HE染色观察其海马形态学变化,免疫组织化学法检测其Bcl-2、Bax蛋白的表达。结果 A组、B组大鼠海马形态学未见明显改变。C组可见海马区细胞肿胀,凋亡细胞增多,细胞层数减少。D组可见大量凋亡细胞,但少于C组,细胞层数较C组增多。A组、B组各时间点Bcl-2、Bax比较均无统计学差异(Pa>0.05)。9 d、15 d、21 d,C组、D组Bcl-2阳性细胞数表达比A组、B组增多,差异均有统计学意义(Pa<0.05)。C组Bax在9 d、15 d分别高于其他3组,差异有统计学意义(Pa<0.05)。D组Bax阳性细胞数表达在9 d时比A组、B组增多(Pa<0.05),但少于C组(P<0.05)。结论 PTZ致可导致大鼠海马区Bcl-2高表达,并持续较长时间,苯巴比妥不能提高Bcl-2的表达。PTZ致后可导致Bax的高表达,但持续时间较短,苯巴比妥可显著抑制Bax的高表达。Objective To explore the effect of phenobarbital on the changes of morphology and Bcl-2 and Bax expression in hippocampus of epileptic rats.Methods Totally 84 SD rats around age of 4 weeks were randomly divided into 4 groups:group A was given normal saline(n=18),group B was given phenobarbital(n=18),group C and group D were epileptic groups(24 rats in each group),of which group C was given normal saline after pentylenetetrazol(PTZ),while group D was given phenobarbital after PTZ.A dose of PTZ 60 mg·kg-1·d-1 was intraperitoneally injected into the rats in group C and group D for 5 days contiguously to induce epilepsy.Since the 6th day,the rats in group A and group C were administered intraperitoneally with physiological saline;the rats in group B and group D were administered intraperitoneally with phenobarbital 50 mg·kg-1·d-1.Paraffin sections were stained with HE stain for morphological evaluation,immunohistochemistry technique was used to investigate the expression of the apoptosis regulatory gene Bcl-2 and Bax,on the 9th,the 15th,and the 21st day.Results HE stains consequence:there was no statistical significance between group A and group B of the changes of morphology.Cell swelling,apoptosis cells and death cells increased while cell layers decreased in hippocampus in group C.Compared with group C,there was less apoptosis cell population in group D,but the layers of the cells were increased.Immunohistochemical staining:there were no significant differences at different time points between group A and group B in the expression of Bcl-2 and Bax protein(Pa0.05).There were more expressions of the Bcl-2 on the 9th,15th and 21st day in group C and group D,and it had statistical significance compared with group A and group B(Pa0.05).There were more expression of the Bax on the 9th and 21st day in group C and there was statistical significances compared with the other 3 groups(Pa0.05),as the expression of the Bax in group D increased on 9th day than that in group A and group B(
