摘  要:目的采用氧糖剥夺预处理模拟脑缺血预处理,探讨预处理对大鼠脑微血管内皮细胞(BMECs)紧密连接的影响。方法体外培养新生大鼠BMECs,给予细胞氧糖剥夺2.5h,再给氧糖3.0h,模拟在体缺血再灌注损伤,以0.5h氧糖剥夺作为预处理。观察细胞超微结构的改变,应用荧光检测细胞间紧密连接相关蛋白ZO-1和细胞骨架蛋白F-actin的位置变化。结果氧糖剥夺可破坏细胞紧密连接等超微结构,使BMECsZO-1和F-actin的膜定位发生改变。预处理可对内皮细胞起到明显的保护作用,稳定ZO-1和F-actin的膜定位。结论预处理可抑制BMECs紧密连接及相关蛋白的改变,对血脑屏障起保护作用。[Objective]To study the effects of preconditioning on tight junctions of brain microvascular endothelial cells, using oxygen glucose deprivation (OGD) to mimic ischemia in vivo. [Methods]Primary culturing the rat brain microvascular endothelial cells, were given 2.5 h OGD and 3 h re-oxygenation/glucose to mimic ischemiareperfusion in vivo with 0.5 h OGD as preconditioning. The changes of cell ultrastructure were tested, and the expression of tight junction proteins ZO-1 and F-actin was assessed by immunofluorescence and fluorescence-labeled assays. [Results]OGD could destroy the cell tight junction ultrastructure, and change the membrane location of ZO-1 and F-actin. OGD preconditioning could significantly alleviate the injury of brain microvascular endothelial cells, and lighten the changes of tight junction proteins ZO-1 and F-actin location. [Conclusion]OGD preconditioning could inhibit the changes of brain microvascular endothelial cells tight junctions and provide protective effects on blood brain barrier.
