摘 要:在单因素实验确定显著因素的基础上,采用响应面法优化气单胞菌(Aeromonas sp.)RS01的发酵培养基。单因素实验确定碳源玉米生淀粉浓度为40g/L,氮源牛肉膏的浓度为12g/L,氯化钠的浓度为3g/L。响应面法优化得到玉米生淀粉、牛肉膏、氯化钠3个显著因素的最佳浓度分别为43.21g/L、12.01g/L、3.21g/L,此条件下低温生淀粉糖化酶酶活达到98.42U/mL,比优化前提高了51.67U/mL,与模型预测值100.32U/mL基本吻合。牛肉膏与NaCl交互作用不显著,玉米生淀粉与NaCl、玉米生淀粉与牛肉膏交互作用显著。The fermentation medium of Aeromonas sp. RS01 was optimized by response surface method after the significant factors were confirmed by single factor experiment. According to single factor experiment, the raw cornstarch as carbon source was 40 g/L, the beef extract as nitrogen source was 12 g/L and the NaC1 was 3 g/L in fermentation medium. The optimum concentrations of raw cornstarch, beef extract and NaCI were confirmed as 43.21 g/L, 12.01 g/L, 3.21 g/L, respectively, by response surface method. Under above conditions the activity of cold-active raw starch-digesting glucoamylase was 98.42U/mL, which was 51.67 U/ml higher than that before optimization and was basically according with the model prediction value of 100.32 U/ml. The interaction between beef extract and NaCl was not significant and those between raw cornstarch and NaCI, beef extract were significant.
