摘  要:建立一种用谷氨酸脱氢酶测定蛋白质的方法。方法:将蛋白质消化成铵,再用谷氨酸脱氢酶动力法在自动生化分析仪上测定铵浓度,计算蛋白质含量。结果:该法回收率为96.6%~103.0%,平均回收率为99.1%,RSD为1.6%。结论:本法准确,精密,抗干扰能力强,且简便快速,与蒸馏法相关性好。OBJECTIVE:To establish a method for assay of protein by glutamate dehydrogenase.METHODS:The protein was completely.degraded to the NH + 4 according to the Kjeldahel procedure,the NH + 4 concentration in the digest juice was measured by glutamate dehydrogenase on automatic analyzer, and the protein concentration was calculated with it.RESULTS:The recoveries ranged from 96.6% to 103.0% , the average was 99.1% . The within run coefficient of variation was 1.6% .Comparing this method with the distillation of the Kjeldahel method,the accuracy was no significantly different( P > 0.05 ),and the coefficient of relativity was 0.999 9 .CONCLUSIONS:This method is simple, sensitive and accurate.This procedure has the advantage of eliminating the distillation and titration steps of the Kjeldahel method and it is ideal for protein assays,since many samples can be run in a single day.
