摘  要:建立反相高效液相色谱检测浓缩苹果汁中的多酚物质如二羟基苯甲酸、绿原酸、咖啡酸、对香豆酸、邻香豆酸、阿魏酸、槲皮素、异槲皮素等方法。HPLC条件为:色谱柱Insertsil ODS-3(250mm×4.6mm,5um),流动相为乙酸水溶液和乙腈,柱温35℃,流速0.8mL/min,线性梯度洗脱,DAD检测器波长为280nm。该方法的检出限为0.0012—0.025mg/L,10种酚类物质的加标回收率均大于85%,测定结果的相对标准偏差为0.9%~2.0%(n=5)。A method of reversed - phase high performance liquid chromatography for determination of polyphenols in apple juice concentrated such as 3,4-dihydroxybenzoic acid, chlorogenic acid, caffeic acid, P- coumaric acid, O- comnaric acid, ferullic acid, quecetin dihydrate and isoquecitin was established. The HPLC conditions were as follows: column insertsil ODS -3 (250 mm × 4.6 mm, 5 um) , mobile phase acetic acid - acetonitrile, column temperature 35℃, flow rate 0.8 mL/min, DAD detector with 280 nm wavelength. The detection limit of the method ranged from 0.001 2 to 0.025 mg/L, the recoveries of polyphenols were more than 85% , and the relative standard deviations were 0.9% - 2.0% (n = 5).
