摘  要:目的:研究大蒜素对内毒素作用下体外培养的人牙周膜成纤维细胞(hPDLCs)Ⅰ型胶原合成及胶原分泌的影响,探讨大蒜素在牙周疾病治疗中的作用。方法:建立hPDLCs有限细胞系,用大蒜素作用于第5代hPDLCs,培养液中加入牙龈卟啉单胞菌脂多糖(LPS),采用细胞免疫荧光化学方法检测细胞内Ⅰ型胶原含量,化学比色法测定细胞培养上清液中羟脯氨酸含量。结果:大蒜素可显著降低LPS对hPDLCsⅠ型胶原合成及胶原分泌的抑制作用(P<0.05),且具有剂量依赖效应。结论:大蒜素可降低LPS对hPDLCsⅠ型胶原合成及胶原分泌的抑制作用,在牙周疾病治疗中具有一定意义。Objective:To study the effect of garlicin on the synthesis and the secretion of collagen typeⅠin human periodontal ligament cells in vitro culture with the contribution of endotoxin,and to probe the role of garlicin on the treatment of the periodontal disease.Methods:The limited cloned strain of hPDLCs(human periodontal ligament cells) was setup,then the garlicin was acted on the fifth generation hPDLCs group the lipopolysaccharide(LPS) of the Porphyromonas gingivalis(P.gingivalis) was added into the culture solution,the content of the intracellular collagen type I was detected via the method of the immunofluorescent cytochemical staining and the content of the hydroxyproline in the supernatant of the cell culture medium was detected through the method of the chemical colorimetry.Results:Garlicin in certain dose range could decrease the inhibitory effect of LPS on the synthesis and the secretion of collagen typeⅠ in hPDLCs in the way of dose-dependent effect(P 〈0.05).Conclusion:Garlicin can decrease the inhibitory effect of LPS on the synthesis and the secretion of collagen typeⅠin hPDLCs,providing definite significance for the treatment of periodontal disease.
