摘  要:以壳聚糖为主要材料,制备壳聚糖微囊。微囊直径在0.5~0.8mm间,微囊在pH4~11的缓冲液中稳定,能耐受3000r/min离心30min以上。用牛血清白蛋白和γ-球蛋白测其通透性,表明随着微囊所在溶液pH的升高,微囊通透性降低。壳聚糖的分子量在一定范围内对微囊通透性几乎无影响而脱乙酰度对通透性影响很大。升高壳聚糖的脱乙酰度,微囊通透性明显降低,而其机械强度无明显变化。用该微囊固定化丝孢酵母细胞,结果酶活力回收达80%以上,操作半衰期达2.5~3月。Chitosan microcapsules with the sizes of 0.5~0.8 mm in diameters were prepared. The microcapsules were found to be stable in the pH range of 4-11, and be able to sustain centrifugation at a level at least for 30 min in 3 000 r/min. The permeability of chitosan microcapsule was reduced with increasing the pH of protein solution in which the diffusion experiment was test. The molecular weight of chitosan in the range from 0.9×10 6 to 1.3×10 6 had little effect on microcapsule permeability, but the degree of deacetylation of chitosan played an important role. Increasing the degree of deacetylation of chitosan from 75% to 96.6%,the permeability of chitosan microcapsule was decreased while the mechaniscal strength remained the same.The recovery of activity of encapsulated yeast cell was more than 80%, and the operation half life was 2.5~3.0 months.
