摘 要:目的:研究含CpG序列的寡核苷酸(unmethylated CpG motif containing oligodeoxynucleotide,CpG ODN)对人肺腺上皮细胞β防御素-2(humanβ-defensin2,hBD-2)表达的影响及其作用途径。方法:培养人肺腺上皮细胞A549,用不同浓度的CpG ODN进行干预。用RT-PCR法和Western Blot法检测刺激后A549细胞hBD-2mRNA和蛋白表达变化及细胞核内NF-κB p65蛋白含量的变化。结果:CpG ODN刺激可以促进A549细胞核内NF-κB p65含量增加,活化NF-κB途径,从而诱导hBD-2的表达,与对照组比较P<0.05;用特异性阻断剂阻断NF-κB活化可以部分抑制CpG ODN的诱导作用。结论:CpG ODN可诱导hBD-2在呼吸道上皮细胞的表达,增加机体对外界微生物的抵抗能力,激活NF-κB信号途径可能是其作用机制之一。Objective To investigate the effects of unmethylated CpG motif containing oligodeoxynucleotide (CpG ODN) on the expression of human β-defensin 2 (hBD-2) and NF-κB p65 in lung epithelial cell line A549. Methods A549 cells were stimulated with different concentration of CpG ODN in the presence or absence of PDTC, an inhibitor of NF-κB. The mRNA and protein expression of hBD-2 were determined by RT-PCR and Western Blot, respectively. The expression of NF-κB p65 in nucleus was detected by Western Blot. Results CpG OND significantly enhanced the expression of hBD-2 in A549 cells. The content of NF-κB p65 in nucleus was markedly up-regulated. Inhibited NF-κB by PDTC suppressed CpG ODN-induced hBD-2 expression. Conclusion CpG ODN might effectively up-regulated hBD-2 expression in lung epithelial cells through activated NF-κB.
