人溶菌酶转基因小鼠乳汁的抑菌活性及稳定性研究

摘　　要：目的研究表达人溶菌酶转基因小鼠乳汁的抑菌活性及稳定性。方法取重组人溶菌酶(rhLYZ)表达的母鼠乳汁和人初乳,以平板扩散法检测乳样的抑菌谱。转基因小鼠乳汁的稳定性研究,系通过将乳样置于不同温度的水浴中加热,测定其溶菌活性;将稀释乳样调节到不同的pH值,或者加入不同种类的金属离子,分别测定酸碱度和金属离子对乳汁抑菌活性的影响。结果人溶菌酶转基因小鼠乳汁对多种革兰阳性及阴性细菌具有不同程度的抑制作用,并具有很强的热稳定性、广泛的pH活性范围,金属离子对其活性的影响较小。结论人溶菌酶转基因小鼠乳汁和人初乳有较相似的生物学活性,是动物乳"人乳化"的良好模型。【Objective】To study the anti-bacterial activities and the stabilities of the milk of the human lysozyme transgenic mice.【Methods】Bactericidal spectrum of the milk of transgenic mice and colostrum to bacterial strains and fungal strain were assayed by agar diffusion test.First,the milk samples were subjected to heat treatment at a series of time,then measure the anti-bacteria activity was measured.Then the milk samples were also diluted with solutions of different pH value and incubated at 37 ℃ for 30 min for detection of their pH stability.They were also mixed with solutions containing different ions at the final concentration of 10 mmol/L.【Results】The milk of hLYZ transgenic mice showed almost the same bactericidal spectrum,similar thermal,pH and ions stability to human colostrums.【Conclusion】The milk of hLYZ transgenic mice is a good model for development of human milklike animal milk.
