摘 要:以稻草秸秆粉为碳源,利用绿色木霉和黑曲霉进行固体发酵产纤维素酶的研究。以混合菌接种比例、麦麸与稻草粉质量比、发酵时间和三角瓶装量4因素为考察对象,通过单因素实验和正交实验优化混合菌固体发酵条件。确定该混合菌株的最优产酶条件,结果表明,混合菌最佳固体发酵条件为:绿色木霉和黑曲霉接种比例为1∶1、麦麸与稻草粉质量比为1∶3.0、发酵时间为4 d、装量为50 mL/1000 mL三角瓶。在此最佳发酵条件下,FPA酶活、CMC酶活、β-G酶活分别为5.29 IU/mL、9.33 IU/mL和49.91 IU/mL,分别是单菌发酵的2.28~2.47倍、2.39~2.45倍、1.38~2.09倍。混合菌发酵产生的各酶活性均高于各种菌单独发酵产生的各酶活性。Straw stalk powder was used as carbon source, and cellulose-producing conditions by solid fermentation of A spergillus niger and Trichoderma viride were investigated. The solid fermentation conditions of mixed strains were optimized by single factor experiments and orthogonal experiments in four aspects including the inoculation ratio, the mass ratio of bran and straw powder, fermentation time and the filled content of flask. Finally the optimum solid fermentation conditions were summed up as follows: the inoculation ratio (Trichoderma viride:Aspergillus niger) was 1 : 1, the mass ratio of bran and straw powder was 1:3, fermentation time was 4 d, and the filled content of flask was 50 mL/1000 mL. Under the above conditions, the activity of FPAase, CMCase and β-Gase were 5.29 1U/mL, 9.33 IU/mL and 49.91 IU/mL respectively, which were 2.28-2.47 times, 2.39-2.45 times and 1.38-2.09 times as high as that by single strain fermentation. In conclusion, enzyme activity by mixed strains fermentation was higher than that by single strain fermentation.
