银耳种质资源遗传多样性及酶学特性分析

摘　　要：从不同地区采集引进了银耳菌株共14株,利用ISSR分子标记技术对这14株银耳菌株进行了遗传多样性分析,从75条引物中筛选出14条引物,共获得105条带,其中多态性条带70条,占条带总数的67%。聚类分析结果表明:在相似水平为70%时,14个菌株可以划分为4个类群,且类群的划分与地理来源之间并无明显相关性,菌株间遗传差异较大,遗传多样性较高,表明供试菌株在DNA分子水平上存在比较显著的遗传变异。利用紫外分光光度法测定了纤维素酶活性,并采用SPSS软件进行了酶活力差异显著性分析。结果显示:供试的14株银耳菌株纤维素酶活大小在1.300～0.922间,纤维素酶活性很低,酶活力差异显著,且与地理来源无明显相关性。14 Tremella fuciformis strains were obtained from several different geographic origins.To analyse the genetic diversity of the 14 strains of Tremella fuciformis,75 ISSR primers were screened and 14 of them were selected to amplify the 14 tested strains and a total of 105 fragments were amplified,and among the total 105 amplified fragments 70（67 %） were polymorphic.These 14 tested strains were divided into 4 groups under similarity of 70 %,with no obvious correlation between geographical distribution and groups,by cluster analysis.The results showed that genetic similarity of these tested strains was low and there were obvious genetic variations in DNA level.Ultraviolet Spectrophotometry were used to determine the activity,and then SPSS were used to analyze the significance of difference of the cellulase of the 14 tseted strains of Tremella fuciformis,the results suggested that the activity of the cellulase of the 14 tested strains was between 1.300 and 0.922,the activity was weak,there were statistically significant differences in the activity of the 14 tested strains,with no obvious correlation between the geographical distribution and 14 tested strains.
