摘  要:目的:通过观察大蒜素对人胃癌细胞增殖、凋亡及凋亡过程中Caspases-3和Caspases-8活性的影响,探讨大蒜素诱导凋亡的机制。方法:采用MTT法检测用药后胃肿瘤细胞的增殖;流式细胞仪分析凋亡细胞;用Caspases-3和Caspases-8活性试剂盒检测Caspase变化。结果:大蒜素对胃肿瘤细胞BGC-823、SGC-7901和MKN-28增殖具有明显的抑制作用,72hIC50分别为65.72、83.16和156.23μmol/L;60μmol/L的大蒜素作用24h后,3种胃肿瘤细胞的凋亡率分别为(21.57±0.83)%、(12.31±0.45)%和(10.97±0.54)%,均高于对照组,P<0.01;大蒜素能活化3种胃肿瘤细胞的Caspases-3和活化BGC-823细胞中Caspases-8。结论:大蒜素诱导人胃肿瘤细胞凋亡过程中,Caspases-3的活化起了重要作用。OBJECTIVE:To investigate the changes and significance of Caspases-3 and Caspases-8 activities in diallyl trisulfide (DATS) induced apoptosis of human gastric cancer cells,and explore the apoptosis mechanism promoted by DATS.METHODS:In vitro experiments,MTT assay was used to determine the cell growth inhibition rate.Cell apoptosis analysis was carried out by flow cytometry.The activity of caspase was determined by Caspases-3 and Caspases-8 Colorimetric Assay Kit.RESULTS:Human gastric cancer cell growth was significantly inhibited by DATS.The IC50 of 72 h after treatment by DATS for BGC-823,SGC-7901 and MKN-28 were 65.72,83.16 and 156.23 μmol/L respectively.After treated with 60 μmol/L DATS for 24 h,the apoptosis rates of human gastric cancer cell were (21.57±0.83)%,(12.31±0.45)% and (10.97±0.54)% respectively.The cell apoptosis rates were significantly higher in the experiment group than the control group (P0.01).DATS activated Caspases-3 of human gastric cancer cells and Caspases-8 of BGC-823 cells.CONCLUSION:DATS induces apoptosis in human gastric cancer cells which may be through the activation of Caspases-3 pathway.
