摘  要:香菇多糖是香菇的重要活性成分.将香菇用热水提取多糖后,采用NaOH溶液对水提后的香菇残渣进一步提取,研究了香菇碱溶性多糖的提取工艺.结果表明,香菇碱溶性多糖的最佳提取工艺为:香菇粉经90℃热水提取2次后,残渣加10倍体积4%NaOH溶液,于60℃提取0.5h,提取2次.碱提液经盐酸中和、离心分离,将沉淀水洗、冻干后,得碱提非水溶性多糖;将上清液减压浓缩、乙醇沉淀、真空干燥后,得碱提水溶性多糖.在此条件下,100g香菇粉可得水溶性多糖3.84g,碱提非水溶性多糖7.36g,碱提水溶性多糖18.68g.Lentinan is the important bioactive component of Lentinus edodes.After extracting polysaccharide with hot water, the residue was further extracted with NaOH solution.The extraction method of alkaline soluble polysaccharide in Lentinus edodes was studied. The results showed that the best extraction conditions were as following: powder of Lentinus edodes was extracted twice by hot water at 90 ℃, then the residue was extracted twice by 4% NaOH solution at 60 ℃ for 0.5 h. Alkaline extract was neutralized by hydrochloric acid and centrifugated. The precipitate was washed by water and freeze-dried to obtain the water-dissoluble polysacchafide. The supemate was vacuum condensed, precipitated by ethanol and vacuum-dried to obtain water-soluble polysaccharide. From 100 g powder of Lentinus edodes, 3.84 g water soluble polysaccharide, 7.36 g polysaccharide alkaline-soluble but waterdissoluble and 18.68 g alkaline soluble polysaccharide could be extracted according to the above conditions.
