莪术醇诱导人胃癌BGC-823细胞凋亡与细胞内活性氧增加的实验研究  被引量:8

摘  要:目的:研究莪术醇抑制BGC-823人胃癌细胞增殖、影响细胞凋亡以及细胞内活性氧(reactive oxy-gen species,ROS)水平变化等生物效应。方法:用MTT法、AO/EB双荧光染色及荧光显微镜、FITC-AnnexinV与PI双染法、流式细胞术检测观察各种生物及形态指标。结果:不同浓度莪术醇在不同时间作用BGC-823,可抑制细胞增殖,并呈浓度、时间依赖性;荧光显微镜观察,经莪术醇作用后,实验组细胞核呈固缩状或圆珠状,可见凋亡小体;人胃癌BGC-823细胞经过不同浓度的莪术醇作用后,凋亡细胞比例明显升高,与对照组比较,差异有显著性(P<0.05):不同浓度莪术醇作用人胃癌BGC-823细胞后,细胞内活性氧水平与莪术醇呈浓度依赖性升高,莪术醇作用24、48h后与对照组比较,有显著性差异(P<0.05)。结论:莪术醇可抑制人胃癌BGC-823细胞增殖及诱导细胞凋亡。莪术醇诱导人胃癌BGC-823细胞凋亡与ROS产生增加相关。Objective:To investigate the effect of curcumol on the proliferation,cell apotosis and intracellular reactive oxygen species(ROS) levels and other biological effects of human gastic cancer BGC-823 cells.Methods: The kinds of guide lines in BGC-823 cells were detected by MTT assay,AO/EB,FITC-Annexin V and PI and flow cytometry methods.Results: Curcumol inhibited cell proliferation of human gastric cancer BGC-823 cells(P〈0.05),in a dose-and-time-dependent manner.After curcumol treated,the nuclei showed pyknosis-like or bead-like,apoptotic bodies by fluorescene microscope,the proportion of apoptotic cells increased significantly(P〈0.05).The difference of Ros between control group and treated group,24,48 h treated with carcumol was significant(P〈0.05).Conclusion: Curcumol can inhibit the proliferation and induce the cell apotosis of BGC-823 cells.The effect of curcumol on BGC-823 cells can induce apotosis which related with increased ROS production.
