摘  要:香草醛酸性试剂比色法是检测儿茶素含量最常见方法,但其准确性及实用性一直受到质疑。利用溶剂萃取、TLC和HPLC等方法,对儿茶素-香草醛酸性试剂显色方法的特异性、准确性进行了研究。结果显示,儿茶素类显色反应的灵敏度远高于黄酮醇、二氢黄酮醇和茶皂素类,又由于这些物质在茶叶中的含量很低,因此不会干扰儿茶素总量的测定。通过对显色试剂中酸的种类、酸浓度、显色温度和显色时间的研究,得出儿茶素检测的最佳方法:采用1%香草醛-浓盐酸试剂,在室温下反应5min,以Y(A505)=aX+b公式计算茶树中儿茶素的总含量。Vanillin-acid reagent spectophotometric methods is usually used to detect catechins content,but its accuracy has been questioned.Solvent extraction,TLC and HPLC analysis were adopted to investigate the specificity and accuracy of vanillin-acid reagent reaction.The results showed that it would not interfere with the determination of catechins because the sensitivity of catechins was the highest compared to those substances such as flavonols,flavanonols and tea saponin.Some factors influencing the reactions of vanillin-acid reagent with catechins,including the kinds of acid,acid concentration,reaction temperature and reaction time were investigated.The results suggested that the best method to analyze catechins content should be performed by using 1% vanillin-HCl reagent and reacting for 5 min under room temperature.Catechins content could be calculated with the formula of Y(A505nm)=aX+b.
