摘  要:报道了乳铁蛋白(LF)的提纯方法,放射免疫分析法的建立及其在肺癌诊断中的应用。用改良的萃取法获得相对分子量为75000D的LF,其聚丙烯酰胺凝胶电泳及免疫双扩散结果均呈单一色带,1mL人乳可提取0.542mgLF。放射免疫分析批间及批内变异分别为4.77%和13.59%,回收率97.99%。对36例肺癌、21例肺部感染和4例正常人的支气管冲洗液中乳铁蛋白(BLF)进行测定,结果表明肺癌组明显高于肺部感染组和正常对照组,其中肺腺癌的检出率最高。病理分期表明,BLF与肺癌的早。The purification of lactoferrin(LF) is reported and its radioimmunoassay applied in the diagnosis of lung cancer (LF RIA) is established by means of a modified method of extraction. LF (relative molecular weight 75 000 Dalton) is obtained. The LF present a single band on both PAGE and double immunodiffussion. The yield of LF is 0.542 mg in 1 mL human milk for this method. The coefficients of variation(CV) within a batch and among batches are 4.77% and 13.59% respectively, and the recovery rate amount to 97.99%. The LF in bronchial washing fluid(BLF) is measured in 36 lung cancer cases, 21 pulmonary infection cases and 4 normal controls. The results suggest that the BLF content in the lung cancer group is higher than that in the pulmonary infection group and the normal control group, and the detection rate is the highest in the lung adenocarcinoma. Pathological analysis indicates that BLF is negatively correlated to the early or later lung tumor.
