摘 要:目的:开发并验证一种简单、重现性好的反相高效液相色谱测定婴儿奶粉中胡萝卜素的方法。方法:碱皂化后,用正己烷提取。采用WatersSunFireC18柱分离,柱温30℃,以甲醇∶三氯甲烷∶异丙醇=70∶25∶5为流动相等度洗脱,流速1mL/min,检测波长为450nm。结果:该方法检测限为0.60μg/100g,重复性和重现性的相对偏差分别为1.59%和3.81%,回收率为96.89%。结论:该方法简便,其精度、回收率、灵敏度都很理想,满足日常分析要求。Objective: A simple and reproducible reversed-phase(RP) high-performance liquid chromatography (HPLC) method for determination of β- carotene in infant formulas was developed and validated. Methods: The method involved saponification and a later extraction of β- carotene with n-hexane. Separation was performed on Waters SunFire C18 column at 30 ℃ using an isocratic mobile phase of hexane-chloroform-isopropyl (70∶ 25∶5, v/v/v) and a flow rate of 1 mL/min. Detection was at 450 nm. Results: The detection limits was 0.60 μg/ 100 g, the relative standard deviations of repeatability and reproducibility were 1.59% and 3.81%, respectively and recovery was 96.89% . Conclusion: The method showed desirable values for precision, recovery and sensitivity, and proved very simple for routine analysis work.
