摘 要:目的了解广西境内海产品牡蛎中分离到的携带tdh和trh两种毒力基因的副溶血性弧菌GX21的基因分子特征,分析与各地副溶血性弧菌的相关性,探讨水产品中副溶血性弧菌流行趋势。方法将监测海产品牡蛎中分离到的一株携带两毒素基因的副溶血性弧菌应用双重PCR的方法检测该菌tdh和trh两种毒力基因。把扩增产物切胶纯化后测序,将测序序列在GenBank DNA序列数据库上BLAST比对,分析比较结果。结果两扩增产物tdh-A和trh-A的序列与GenBank数据库中的多个序列有极高的相似度,最高达99%,只有一个碱基的错配或一个gaps。结论扩增产物tdh-A为副溶血性弧菌GX21的tdh基因不完全cds,trh-A为副溶血性弧菌GX21的trh基因不完全cds。比对表明副溶血性弧菌GX21与日本学者在1994年公布的副溶血性弧菌(TH3766)有很高的同源性。Objective To study the molecular characteristics of tdh and trh genes of a Vibrio parahaemolyticus strain,GX21 which was isolated form seafood-oysters in Guangxi and analyze epidemic trend on V.parahaemolyticus in marine products by matching relationship between different strains from the world.Methods Double PCR method was used to amplify the tdh and trh from GX21,then the products of amplification were recovered and sequenced,and analyzed information by matching the DNA sequence of amplification with the BLAST on the NCBI GenBank DNA databases.Results The sequences of tdh-A and trh-A were highly similar to many sequences in GenBank;The highest similarity was 99%,and there was only one base mismatch or one gap.Conclusion The amplification products tdh-A is partial coding region of tdh gene from V.parahaemolyticus strain,GX21;and trhA is partial coding region of trh gene.The V.parahaemolyticus GX21 is highly homologous with V.parahaemolyticus TH3766 which was found by a Japan scholar in 1994.
