摘  要:研究从红薯叶中提取的水溶性多糖的分离、纯化,单糖组成和结构,为红薯叶的开发利用提供参考.通过正丁醇-三氯乙酸(体积比为20∶1)除蛋白、透析法除小分子杂质后,经Seph-adex G-100葡聚糖凝胶柱分离得到了精制多糖IBL(I).通过凝胶过滤法、薄层层析(TLC)、气相色谱(GC)、紫外光谱(UV)、红外光谱(IR)等分析手段初步确定了其组成和结构.红薯叶多糖IBL(I)为均一组分,相对分子质量为2.63×104,其单糖组成为木糖、甘露糖、葡萄糖,其物质的量的比为0.47∶0.35∶0.18.IBL(I)具有典型的多糖特征吸收峰,其结构中存在β型糖苷键.We studied the separation,purification,monosaccharide composition and structure of water-soluble polysaccharides extracted from Ipomoea batatas Lam leaves so as to provide reference for the development of Ipomoea batatas Lam leaves.The method comprised the following steps: removing proteins by using n-butanol and trichloroacetic acid with the volume ratio of 20∶1;removing small-molecule impurities by dialysis;separating refined polysaccharide IBL(I) by Sephadex G-100 gel column chromatography;and determining the composition and the structure of the polysaccharide IBL(I) by gel filtration,thin-layer chromatography,gas chromatography,ultraviolet spectroscopy,infrared spectroscopy and so on.The results show that the polysaccharide IBL(I) has a homogeneous component with the relative molecular weight of 2.63×10^4,and contains xylose,glucose and mannose with the molar ratio of 0.47∶0.35∶0.18;and polysaccharide IBL(I) has a typical characteristic absorption peak,which identifies that the polysaccharide IBL(I) contains a β-glucosidal bond.
