摘  要:目的:检测复方奥硝唑-甲磺酸培氟沙星缓释制剂是否存在潜在的遗传毒性。方法:采用60只昆明小鼠分为6组,每组10只,按10、5、2.5、1.25g/kg经口灌胃给药,取胸骨骨髓进行骨髓微核实验,;组氨酸缺陷型鼠伤寒沙门氏菌TA97、TA98、TA100和TA102菌株用平板掺入法实验。实验将受试药品分为5个剂量(0.001、0.01、0.1、1.0、10μg/皿),在有、无代谢活化剂S9的条件下对药物进行相关检测。结果:小鼠骨髓微核数目未见明显增多,各实验组微核诱发率与阴性对照组比较差异均无统计学意义(P>0.05);TA97、TA98、TA100和TA102菌株回复突变菌落未见明显增多,各实验组与阴性对照组比较(P>0.05)。结论:复方奥硝唑-甲磺酸培氟沙星缓释制剂未见潜在遗传毒性。AIM:To examine whether the"ornidazole and pefloxacinmesylat compound"has potential genotoxicity.METHODS:Sixty Kunming mice were randomly divided into 6 groups.4 groups were orally administered with 10g/k,5g/kg,2.5g/kg,and 1.25g/kg of the compounds respectively.The negative control group was given water and the positive control group was periotoneally injected with 40 mg/kg cyclophosphamide.After 24 h,the animals were sacrificed and the bone marrow of sternum was stained with Giemsa method.The number of micronucleus in polychromatic erythrocytes was calculated.Ames Test was conducted with GB15193,4294 standard.Histidine-defective Salmonella typhimurium TA97,TA98,TA100,and TA102 strains were treated with 0.001μg/dish ~ 10μg/dish of the compounds.After 48 h,the muted colonies were counted.RESULTS:No obvious difference was found between the compound groups and the negative control group in the micronucleus test.Ames test results showed that TA97,TA98,TA100,and TA102 strains had no significant increase in reverse mutation colonies.CONCLUSION:Ornidazole and pefloxacinmesylat compound showed no genotoxicity.
