摘 要:目的探讨维生素A在体内的活性产物视黄酸(RA)对1型糖尿病(T1DM)患儿外周血Th1/Th2平衡的影响。方法选取32例T1DM患儿为T1DM组,28例健康儿童为健康对照组。2组均在无菌条件下获取外周血单个核细胞(PBMC)悬液,均体外培养48 h。ELISA法检测培养上清液中IFN-γ、IL-4水平变化。采用SPSS11.5软件进行统计学分析。结果 1.T1DM组PBMC产生IFN-γ水平高于健康对照组(P<0.01),IFN-γ/IL-4比值明显高于健康对照组(P<0.01),IL-4水平与健康对照组比较差异无统计学意义(P>0.05);2.维生素A干预后T1DM组干预前PBMC产生IFN-γ水平明显低于(P<0.01),IFN-γ/IL-4比值明显低于T1DM组(P<0.01),IL-4水平高于T1DM组,但差异无统计学意义(P>0.05)。结论维生素A主要通过抑制Th1细胞功能,显著改善T1DM患儿Th1/Th2的失衡状态。Objective To explore the influence of retinoic acid(RA),the metabolic product of vitamin A in vivo,on the balance of Th1/Th2 in children with type 1 diabetes mellitus(T1DM).Methods Totally 32 children with T1DM were taken as T1DM group,and 28 healthy children as healthy control group.Peripheral blood mononuclear cells(PBMC) were isolated from venous blood and made into cells suspension.PBMC of T1DM children were divided into 2 groups: T1DM group and vitamin A group.The supernatant was collected after 48 h.The levels of IFN-γ and IL-4 in the supernatant were determined by enzyme-linked immunosorbent assay(ELISA).SPSS 11.5 software was used to analyze the data.Results 1.IFN-γ level produced from PBMC in T1DM group was higher than that in healthy control group(P〈0.01),IFN-γ/IL-4 ratio was higher than that in healthy control group(P〈0.01),and there were no significant differences of the level of IL-4 between the 2 groups(P〈0.05).2.The level of IFN-γ produced from PBMC in vitamin A group was lower than that in T1DM group(P〈0.01),IFN-γ/IL-4 ratio was lower than that in T1DM group(P〈0.01),and the level of IL-4 was higher than that in T1DM group,but the difference was not statistically significant(P〈0.05).Conclusion Through inhibiting the function of Th1,vitamin A can restore the balance of Th1/Th2 in children with T1DM.
