尿激酶型纤溶酶原激活剂及其抑制剂PAI-1 mRNAs的表达与人肝细胞癌的关系

摘　　要：为探讨尿激酶型纤溶酶原激活剂（ｕＰＡ）及其抑制剂ＰＡＩ－１ｍＲＮＡｓ的表达变化与人肝细胞癌（ＨＣＣ）浸润、转移间的关系，应用Ｎｏｒｔｈｅｒｎ印迹杂交技术对３０例新鲜活检肝组织作ｕＰＡ和ＰＡＩ－１ｍＲＮＡｓ表达的研究。结果显示：１６例伴肝内浸润和转移癌组织中ｕＰＡ和ＰＡＩ－１ｍＲＮＡｓ表达水平明显高于１４例无肝内浸润和转移肝组织；１８例高分化ＨＣＣ癌组织与１２例中、低分化ＨＣＣ相比，ｕＰＡ和ＰＡＩ－１ｍＲＮＡｓ含量差异有显著性意义。结果表明，癌组织内ｕＰＡ和ＰＡＩ－１基因转录水平的改变与ＨＣＣ浸润、转移及细胞分化密切相关。To study the relationship between the changes in urokinase molecular system and human hepatocellular carcinoma, the expression of urokinase type plasminogen activator (uPA) and its inhibitor type 1 (PAI 1) mRNAs was analysed by using Northern blot method in 30 biopsied HCC tissues with intrahepatic invasion and metastasis and those without invasion and metastasis. Increased expression of uPA and PAI 1 mRNAs was noted in poorly differentiated HCC when compared with that in well differentiated HCC. It is concluded that the changes in expression of uPA and PAI 1 mRNAs may be related to HCC invasion, metastasis and cell differentiation.
