摘　　要：目的 探讨分离胃黏膜上皮细胞的方法及对其细胞膜蛋白质的初步分析.方法 采用0.1%的胃蛋白酶溶液消化掉附在胃黏膜上的黏液,刮取黏膜,用机械法将黏膜剪碎,过滤,离心,从而获得胃黏膜上皮细胞 用Triton-114方法提取细胞质膜蛋白质,SDS-PAGE电泳,并对其中的几个条带质谱分析.结果 成功获得了胃黏膜上皮细胞 胃黏膜上皮细胞质膜蛋白质得到了较好的分离 SDS-PAGE电泳细胞膜蛋白质得到了清晰的条带 用质谱分析条带获得了一些细胞膜蛋白信息.结论 利用物理和化学相结合的方法可以分离得到胃黏膜上皮细胞,成功提取胃黏膜上皮细胞质膜蛋白质并进行了初步鉴定.Objective To explore the method of separating gastric epithelium mucosae cells and analyze the cell membrane proteins. Methods Get rid of the mucus of the gastric tissues by 0. 1% pepsin solution, scrape the mucous membrane, shear the tissues by mechanical method and then after filtering and centrifugation, the gastric epithelium mucosae cells was obtained. Extract membrane proteins by Triton-114,perform SDS-PAGE electrophoresis and choose some protein straps to do mass spectrography analysis. Results We obtained gastric epithelium mucosae cells successfully,the cell plasma membrane proteins were separated from total proteins, and we acquired some membrane proteins information by mass spectrography analysis. Conclusion Gastric epithelium mucosae cells can be separated by the combination of physical and chemical methods,plasma membrane proteins can be extracted by lyses buffer containing Triton-114.
