壳寡糖对急性酒精性肝损伤保护作用的研究

摘　　要：目的探讨壳寡糖对急性酒精性肝损伤的保护作用。方法30只大鼠随机分成酒精组(10只),壳寡糖组(10只),对照组(10只)。酒精组大鼠按体重15g/kg一次性灌胃47.5%酒精,壳寡糖组大鼠按体重0.01mL/g灌胃壳寡糖溶液(0.67g/mL)后0.5h再按体重15g/kg一次性灌胃47.5%酒精。对照组大鼠灌胃生理盐水。灌胃后48h取血清检测ALT、AST和GGT,取肝组织做病检和TUNEL检测。结果壳寡糖组ALT较酒精组极显著降低,P<0.01。酒精组大鼠肝组织水变性、脂肪变性、片状坏死和细胞凋亡病变率均为100%;壳寡糖组大鼠肝组织水变性和细胞凋亡病变率均为100%,无脂肪变性、片状坏死。酒精组凋亡肝细胞核数占肝细胞核总数比是48.2%,壳寡糖组是26.7%,P<0.01。对照组未见异常病变。结论壳寡糖对酒精性肝损伤具有一定的保护作用。【Objective】To study the protective effect of chitooligomer to the alcohol-induced liver injury.【Methods】30 rats were randomly divided into the alcohol group with 10 rats,the chitooligomer group with 10 rats and the control group with 10 rats.The alcohol group was given the alcohol（47.5% v/v） at the dose of 15 g/kg body weight by gavages,and the chitooligomer group was given the alcohol（47.5% v/v） at the dose of 15 g/kg body weight after 0.5 h when the rats were given the solution of chitooligomer（0.67 g/mL） at the dose of 0.01 mL/g body weight by gavages,and the control group was given the physiological saline.These rats were killed after 48 h.Their bloods were drawn for the detection on ALT,AST and GGT,and their hepatic tissues were examined by light microscope,TUNEL.【Results】ALT of the chitooligomer group is lower than the alcohol group,P〈0.01.The rates of the hydropic degeneration,fatty degeneration,patchy necrosis,and apoptosis cells in hepatic tissues in the alcohol group were all 100%.The rates of the hydropic degeneration and apoptosis cells in hepatic tissues in the chitooligomer group were both 100%,but the fatty degeneration and patchy necrosis were not found.The rate of hepatocyte nuclei with positive apoptosis was 48.2% in the total hepatocyte nuclei in the alcohol group,but the chitooligomer group was 26.7%,P〈0.01.No abnormity was found in the physiological saline control group.【Conclusion】Chitooligomer has protective effect on alcohol-induced liver injury.
