摘  要:探讨自然光制下正常成年人外周血淋巴细胞钟基因Clock和Bmal1的昼夜节律性表达。用实时定量RT-PCR方法,测定不同昼夜时点(ZT)受试者外周血淋巴细胞总RNA中核心钟基因Clock和Bmal1的mRNA表达量,通过余弦法和Clock Lab软件获取节律参数,并经振幅检验分析是否存在昼夜节律。结果发现正常成年人外周血淋巴细胞钟基因Clock和Bmal1的mRNA表达呈昼夜节律性振荡(P<0.05),Clock的峰时和谷时分别位于ZT13和ZT1,Bmal1的峰时和谷时分别位于ZT12和ZT24;两个基因在所检测的各个昼夜时点中表达水平均有明显差异(P<0.05),Bmal1的表达水平较Clock降低;二者表达的峰值相位、振幅、峰时和谷时相一致(P>0.05),但Bmal1转录的中值水平以及峰时mRNA水平和谷时mRNA水平降低(P<0.05)。提示正常成年人外周血淋巴细胞钟基因Clock和Bmal1的表达存在同步化的昼夜节律性转录特征。This study was conducted to investigate the circadian expressions of core clock genes,Clock and Bmal1 in the peripheral blood lymphocytes of healthy young adults.Lymphocytes were separated from blood and the total RNA was extracted from each sample.The quantitative real time reverse transcription polymerase chain reaction(RT-PCR) was used to determine the temporal changes in mRNA levels of Clock and Bmal1 during different zeitgeber times(ZT).The circadian parameters were obtained and analyzed by the cosine function,Clock Lab software and the amplitude F test to reveal the genes' circadian rhythm in the LD(16:8) condition.It was found that both the Clock and Bmal1 mRNA levels in the human peripheral lymphocytes showed a significantly circadian oscillation(amplitude F test,P 0.05),with the peak at ZT13,trough during ZT1 for the Clock,and the peak during ZT12,trough at ZT24 for the Bmal1.In comparison with the Clock,the expressive levels at different time points,mesor,mRNA level at peak and at trough of the Bmal1 in the adult human peripheral lymphocytes were correspondingly smaller(P0.05) but parameters such as peak phase,amplitude,peak time and trough time of the Bmal1 expression were similar to those of the Clock(P0.05).These findings suggest that the expressions of Clock and Bmal1 in the human peripheral blood lymphocytes display a synchronous circadian transcription characteristics.
