摘  要:目的检测促血管生成素1,2(angiopoietin1,2)及其受体Tie-2在大肠癌(colorectal carcino-ma)组织中的表达,探讨其与肿瘤血管形成的关系。方法采用免疫组织化学法(PV-9000)和半定量逆转录聚合酶链反应(RT-PCR)法检测大肠癌和癌旁正常组织Ang-1,Ang-2及Tie-2的表达,应用CD105检测组织微血管密度(microvessel density,MVD)。结果 Ang-1、Ang-2及其受体Tie-2在正常大肠黏膜组织和大肠癌组织中均有表达,大肠癌组织中Ang-2、Tie-2蛋白表达的阳性率明显高于正常大肠组织,差异有统计学意义(P<0.05);Ang-1在大肠癌中的mRNA表达低于癌旁正常组织,但差异无统计学意义(P>0.05),Ang-2在大肠癌组织mRNA表达明显高于癌旁正常组织,差异有统计学意义(P<0.05),Tie-2在大肠癌中的mRNA表达略高于癌旁正常组织,差异无统计学意义(P>0.05)。结论 Ang-2在大肠癌的血管生成过程中起着重要的作用,与肿瘤的发生发展密切相关。Objective To explore angiopoietin-1,2 and their receptor tie-2 expression and their significance in tumor angiogenesis in colorectal carcinoma.Methods The expression of angiopoietin-1,2 and their receptor Tie-2 in the colorectal carcinoma were detected by reverse transcription-PCR (RT-PCR) and immunohistochemical PV-9000 two steps methods,and to detect the microvessel density staining with CD105 also by immunohistochemical PV-9000 two steps method.Results Expression of angiopoietin-1,2 and their receptor Tie-2 were detected in both normal colorectal tissues and colorectal carcinoma.The protein expression level of Ang-2 and Tie-2 in colorectal carcinoma was higher than those in normal colorectal tissues (P0.05),but Ang-1 expression is in reverse.Ang-2 mRNA expression in colorectal carcinoma (0.50±0.09) was higher than that in paraneoplastic tissues(P0.05).Conclusion Ang-2 may play a very important role in the development of colorectal carcinoma and is closely correlated with angiogenesis.
