摘 要:对采自武汉东湖水华蓝藻细胞中的微囊藻毒素(Microcystins,MC)的提取方法进行了研究,比较了不同提取溶剂对MC的提取效果。结果表明,5%乙酸溶液和连续抽提法(先用5%乙酸溶液,然后用40%甲醇溶液)能最大限度地从藻细胞中分别提取MC-RR和MC-LR,而且其提取液pH值接近藻胆蛋白的等电点,可省去调pH值法去除藻胆蛋白的步骤,使整个提取过程更加简捷、高效。正丁醇∶甲醇∶水(5∶20∶75,体积比)和甲醇溶液(40%、80%和100%)对MC的提取效率均远远低于5%乙酸溶液和连续抽提法。此外,提取时间的长短也直接影响提取液中MC的浓度。This paper investigated the extraction of microcystins from natural cyanobacterial bloom taken from the East Lake in Wuhan city.The extraction methods were assessed with variant solvents,and 5% acetic acid was found to be an optimum solvent for extracting MC-RR,the continuously extraction method could extract more MC-LR from cyanobacterial cells than the others.Futhurmore,the pH values of the extracts obtained by above two methods were close to the isoelectric point of phycobiliproteins,hence the adjustement of pH value to remove phycobiliproteins could be omitted,which made the extraction process more simple with higher efficiency.Butanol-methanol-water(5∶20∶75)and methanol aqueous(40%,80% and 100%)had far less efficiency than 5% acetic acid and the continuously extraction method.In addition,extraction time also influenced the extraction efficiency of microcystins.
