多肋藻(Costaria costata)多糖的提取分离及理化性质分析

摘　　要：目的从多肋藻(Costaria costata)中提取分离多糖并对其进行理化性质分析。方法采用水和2%碳酸钠水溶液于85℃提取多糖,采用高效凝胶渗透色谱法(HPGPC)分析多糖相对分子质量,用核磁共振氢谱(1H-NMR)法分析褐藻胶中M/G比,用高效液相色谱法(HPLC)分析褐藻糖胶单糖组成。结果从多肋藻中得到1种褐藻胶(ALG)和2种褐藻糖胶(CWF,CAF),其得率分别为42.4%、3.32%和3.52%;相对分子质量分别为212、335和195kD;ALG中M/G比为1.55;CWF和CAF中均含有岩藻糖、半乳糖、甘露糖、葡萄糖醛酸、鼠李糖、木糖和葡萄糖胺等7种单糖,其摩尔比分别为50.54、20.53、10.90、8.87、2.67、4.30、2.20及47.52、32.10、7.90、4.28、3.70、1.20、3.30;FTIR显示CWF和CAF中含有硫酸酯基,经分析表明其含量分别为23%和9%。结论多肋藻中多糖主要含有褐藻胶以及2种结构复杂的褐藻糖胶。Objective To extract polysaccharides from Costaria costata and analyse their physicochemical characters.Methods The polysaccharides were extracted with water and 2% sodium carbonate water solution at 85 ℃,their relative molecular masses（Mr） were analyzed by high performance gel permeation chromatography（HPGPC）,the M/G ratio of alginate was determined by 1H-NMR,and the monosaccharides composition of fucoidans were determined by high performance liquid chromatography（HPLC）.Results The polysaccharides of alginate（ALG） and two kinds of fucoidan（CWF,CAF） were extracted from C.costata with the yield of 42.4%,3.32% and 3.52%,respectively.Their Mr were 212,335 and 195 kD,respectively.The M/G ratio of ALG was 1.55.There were seven kinds of monosaccharides in CWF and CAF,and the molar ratio of fucose,galactose,mannose,glucuronic acid,rhamanose,xylose and glucosamine were 50.54,20.53,10.90,8.87,2.67,4.30,2.20 and 47.52,32.10,7.90,4.28,3.70,1.20,3.30,respectively.The FTIR spectra,both CWF and CAF showed specific absorption at 1235 cm-1,which indicate the existance of sulfate in the polysaccharides,and the sulfate contents of CWF and CAF were 23% and 9%,respectively.Conclusion The polysaccharides in C.costata were mainly composed by alginate and two kinds of structural complex fucoidans.
