摘  要:目的:观察复方奥硝唑甲磺酸培氟沙星牙周缓释制剂对大鼠睾丸超微结构和氧化损伤指标的影响。方法:40只SD雄性大鼠按随机数字表法分为阴性对照组和低、中、高剂量组,每组10只;低、中、高剂量组每日按体质量(1、4、8 g/kg)灌服复方奥硝唑甲磺酸培氟沙星牙周缓释制剂,阴性对照组每日按体质量灌服双蒸水,连续42 d,与雌鼠交配成功后处死雄鼠,计算睾丸、附睾系数,进行精子计数和活动度检查,睾丸行超微结构观察,并检测睾丸中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力。结果:高剂量组睾丸、附睾系数减小,精子数量减少、活动度降低,与阴性对照组比较均有统计学意义(P<0.05),高剂量组睾丸超微结构有损伤;中剂量组睾丸MDA含量上升、CuZn-SOD活力降低,高剂量组睾丸MDA含量上升、Mn-SOD活力和CuZn-SOD活力下降,与阴性对照组比较均有统计学意义(P<0.05)。结论:高剂量复方奥硝唑甲磺酸培氟沙星牙周缓释制剂对大鼠睾丸有一定的毒性,MDA含量升高、SOD活力下降可能是其作用机制之一。AIM: To investigate the testicular toxicity of a ornidazole and pefloxacin mesylate containing,sustained-release periodontal compound on male rats.METHODS:A total of 40 male SD rats were randomly divided into negative control,low-,medium-and high-dose groups by random digits table,with 10 animals in each group.The rats of low-,medium-and high-dose groups were fed daily with the sustained release drug at 1,4,and 8g/kg respectively,and those of the negative group were fed daily with distilled water,for 42 consecutive days.Then all rats were killed after success mating.The coefficient of testis and epididymis,sperm count and movement,testicular ultrastructure,malondialchehyche(MDA) content and superoxide dismutase(SOD) activity in testis were determined.RESULTS:Coefficient of testis and epididymis,sperm counting and sperm mobility were significantly reduced in high-dose group(P0.05).In addition,ultrastructural damage was observed.In both medium-dose and high-dose groups,MDA content increase and CuZn-SOD activity decrease were significant(P0.05).But significant Mn-SOD activity decrease was detected only in high-dose group(P0.05).CONCLUSION:The sustained-release,ornidazole and pefloxacin mesylate containing,periodontal compound has certain testis toxicity.The underlying mechanism might be the increased MDA content and decreased SOD activity.
