NDPK/nm23基因在滋养细胞肿瘤中的表达及其意义

摘　　要：目的：探讨ｎｍ２３基因表达产物（ＮＤＰＫ）在滋养细胞中的表达及其意义。方法：采用免疫组织化学方法，检测经病理证实的正常绒毛（２５例）、葡萄胎（３０例）、侵蚀性葡萄胎（２５例）、绒毛膜癌（２６例）组织中ＮＤＰＫ／ｎｍ２３的表达状况。结果：①５１例滋养细胞肿瘤中ＮＤＰＫ／ｎｍ２３的阳性表达率为１５．７％（８／１５），远低于正常绒毛组织７６．０％（１９／２５），差异有高度显著性（Ｐ＜０．０１）。②葡萄胎、侵蚀性葡萄胎和绒毛膜癌中ＮＤＰＫ／ｎｍ２３的阳性表达率分别为６６．７％、２８．０％、３．９％，差异有显著性（Ｐ＜００５）。③伴有转移的１４例滋养细胞肿瘤中，无１例ＮＤＰＫ／ｎｍ２３阳性表达。④ＮＤＰＫ／ｎｍ２３在葡萄胎中阳性表达率稍低于正常绒毛对照组，差异无显著性（Ｐ＞００５）。结论：ＮＤＰＫ／ｎｍ２３的低表达或不表达与滋养细胞肿瘤的发生、转移、恶性程度及预后密切相关。Objective: To study the expression and significance of nucleoside diphosphate kinase(NDPK), the product of metastasis suppressor gene nm23, in gestational trophoblastic tumor. Methods: The expression of NDPK/nm23 was determined from formalin fixed, paraffin embedded tissues of 25 normal chorion tissue, 30 hydatidiform mole, 51 gestational trophoblastic tumor(including 25 invasive hydatidiform mole, 26 choriocarcinoma), using an immunohistochemical technique. Results: (1) The possitive rate of NDPK/nm23 gene expression in gestational trophoblastic tumor was 15.7%(8/51), significantly lower than the normal group( P <0.01). (2) There was a gradual decrease of gene NDPK/nm23 expression among hydatidiform mole, invasive hydatidiform mole and choriocarcinoma significantly. (3) There was not a single expression of NDPK/nm23 in gestational trophoblastic tumor cases with metastasis(14 cases). (4) NDPK/nm23 expression rate in hydatidiform mole(66.7%), slightly lower than normal chorion tissue(with a positive expression rate of 76.0%), but that difference was not significant( P >0.05). Conclusion: lower or lack of expression of NDPK/nm23 gene appear to be ralated to the genesis, metastasis, the degree of malignancy and prognosis of gestational trophoblastic tumor.
